Sensory gating, MMN, and P300 have been demonstrated to be impaired in subjects clinically at risk of developing a psychotic disorder. In the meta-analysis, duration MMN achieved the highest effect size measures.


In summary, MMN studies have produced the most convincing results until now, including independent replication of the predictive validity. However, a synopsis of the literature revealed a relative paucity of ERP studies addressing the psychosis risk state. Considering the high clinical relevance of valid psychosis prediction, future research should question for the most informative paradigms and should allow for meta-analytic evaluation with regard to specificity and sensitivity of the most appropriate parameters.

ABSTRACT Background: Individuals with an “Attenuated Psychosis Syndrome” (APS) have a 20–40% chance of developing a psychotic disorder within two years; however it is difficult to predict which of them will become ill on the basis of their clinical symptoms alone. We examined whether P50 gating deficits could help to discriminate individuals with APS and also those who are particularly likely to make a transition to psychosis. Method: 36 cases meeting PACE (Personal Assessment and Crisis Evaluation) criteria for the APS, all free of anti-psychotics, and 60 controls performed an auditory conditioning–testing experiment while their electroenceph-alogram was recorded. The P50 ratio and its C–T difference were compared between groups. Subjects received follow-up for up to 2 years to determine their clinical outcome. (繼續閱讀…)

Frontal P3 Event-Related Potential is related to Brain Glutamine/Glutamate Ratio Measured In Vivo.

ABSTRACT The auditory P3 event-related potential (ERP) is thought to index cognitive processing relevant to attention and working memory processes. Drug challenge studies suggest that glutamate neurotransmission plays an important role in modulating P3 ERP. However, while direct links between glutamate activity and P3 ERP response in humans are suspected, mechanistic details remain largely unknown. We investigated here the relationships between P3 ERP and indices of glutamatergic processing measured in vivo with proton magnetic resonance spectroscopy ((1)H MRS).

We hypothesized that a higher index of glutamatergic processing (Glutamine/Glutamate ratio; abbreviated Gln/Glu) in the anterior cingulate (ACC) and in the parietal-occipital (POC) cortices would associate with larger frontal P3a and parietal P3b amplitudes, respectively. (繼續閱讀…)

Δ(9)-THC Disrupts Gamma (γ)-Band Neural Oscillations in Humans.

Gamma (γ)-band oscillations play a key role in perception, associative learning, and conscious awareness and have been shown to be disrupted by cannabinoids in animal studies. The goal of this study was to determine whether cannabinoids disrupt γ oscillations in humans and whether these effects relate to their psychosis-relevant behavioral effects. The acute, dose-related effects of delta-9-tetrahydrocannabinol (Δ(9)-THC) on the auditory steady-state response (ASSR) were studied in humans (n=20) who completed three test days during which they received intravenous Δ(9)-THC (placebo, 0.015 and 0.03 mg/kg) in a double-blind, randomized, cross-over, and counterbalanced design. Electroencephalography (EEG) was recorded while subjects listened to auditory click trains presented at 20, 30 and 40 Hz. January 14, 2015於JAMA Psychiatry上線的新文章Spontaneous Gamma Activity in Schizophrenia,作者群是 Yoji Hirano; Naoya Oribe等日本團隊,但是指導者是 Kevin M. Spencer  24個控制組與24個思覺失調組進行ASSR

This cross-sectional study reports an increase in spontaneous EEG gamma activity during auditory steady-state stimulation in schizophrenia, reflecting a disruption in the normal balance of excitation and inhibition.




提到MMN,大家必然想到 discovered in 1978 by 芬蘭籍Risto Näätänen,但如果侷限於美國學者,Daniel C. Javitt 是美國MMN第一把交椅。

P50呢? 擔任過  Am J Psychiatry 總編輯的  Robert Freedman,  是全球P50 第一把交椅。

可想而知:美國的MMN大老與P50大老聯合撰寫之Review 有多麼值得一讀: 如果今年你只想看一篇ERP的文章,就看這一篇吧^_^

Sensory Processing Dysfunction in the Personal Experience and Neuronal Machinery of Schizophrenia   Am J Psychiatry 172:1, January 2015

Sensory processing deficits, first investigated by Kraepelin and Bleuler as possible pathophysiological mechanisms in schizophrenia, are now being recharacterized in the context of our current understanding of the molecular and neuro- biological brain mechanisms involved. The National Institute of Mental Health Research Domain Criteria position these deficits as intermediaries between molecular and cellular mechanisms and clinical symptoms of schizophrenia, such as hallucinations. The prepulse inhibition of startle responses by a weaker preceding tone, the inhibitory gating of re- sponse to paired sensory stimuli characterized using the auditory P50 evoked response, and the detection of slight deviations in patterns of sensory stimulation eliciting the cortical mismatch negativity potential demonstrate deficits in early sensory processing mechanisms, whose molecular and neurobiological bases are increasingly well understood. Deficits in sensory processing underlie more complex cog- nitive dysfunction and are in turn affected by higher-level cognitive difficulties. These deficits are now being used to identify genes involved in familial transmission of schizo- phrenia and to monitor potentially therapeutic drug effects for both treatment and prevention. This research also pro- vides a clinical reminder that patients’ sensory perception of the surrounding world, even during treatment sessions, may differ considerably from others’ perceptions. A person’s ability to understand and interact effectively with the sur- rounding world ultimately depends on an underlying sensory experience of it.
